The PureEmbryo Series

Development in progress.

Although Nidacon has concentrated on improved function and production .of products for the preparation of human sperm, the embryologists and biochemists in our research and development group have been quietly collecting all new data concerning media for the in vitro culture of the pre-embryo. During this work over the last few years, many new ideas have been incorporated. These ideas have led to considerable changes in the standard media. Each and every new character has been individually tested in a number of ways, simplex optimization. It is these new ideas that should provide us with better tools for helping patients, taking into account the very special cell physiology. A number of our ideas have never previously been used in any embryo culture medium.

– The embryo is developing in the maternal environment during its travel to the uterus. Consequently, one must take into account the composition of this maternal environment in order to provide the most correct conditions for in-vitro embryo development. During the 4-5-day travel of the embryo, it is under the influence of the maternal hormones during that period of the menstrual cycle. Two important hormones involved are estrogen (E2) and progesterone (P4), and it is known that they have an effect on development, among other things, by the fact that human embryos have receptors for E2 and P4.

- Well known in the endocrinological control of cell growth is that the cells need to take up carbohydrates and protein from their environment, the building blocks cell growth. Carbohydrate and protein transport across cell membranes and into cells is facilitated by insulin. The lack of insulin in the human embryo culture medium has a detrimental effect on the cell growth and, therefore, needs to be present in the medium to replace that supplied by the maternal organism.

- A change to a more physiological pH in culture medium could improve today’s results. Studies done by Paul Holmes have shown that the pH in today’s media does not correspond with pH in the uterus. This data has also been verified by others.

Human clinical trials were started some months ago and they look very promising. It seems that our efforts will soon result in something extra for the market. However, we are not going to rush this very important development, especially since human children and families are involved. We need very good, reassuring results before we proceed through the details of registrations with medical authorities. It is also quite important for us, for our distributors and for our clients to incorporate a long shelf into these media products; this has been a continual goal.

The entire research and development process has been, and still is, a very fascinating journey and we’re getting closer to our goals every day, a series of media including everything needed by the sperm, the oocytes and the pre-embryos.

The PureEmbryo Series

PureFlush PureFlush – pH stable media to be used during aspiration of the oocytes and for work outside the incubator. Containing antioxidants for protection against reactive oxygen species (ROS) . The antioxidants have been chosen in collaboration with the expert Dr Elkin Lucena at Cecolfes, Centro Colombiano de Fertilidad y Esterilidad in Bogota, Colombia.

PureFert PureFert – media for fertilisation using standard IVF. Optimised to provide the best environment for both sperm and oocyte. A stable salt balance, carbohydrates for energy , the amino acids needed and reaction cofactors.

PureCleave PureCleave – media for culturing embryos after fertilisation up to 8-cell stage on day 3. Contains an optimised balance of salts, amino acids, hormones and vitamins in order to give the embryo the best possible environment.

PureBlast PureBlast media for culturing embryos from day 3 (6-8 cells) to day 5-6 (blastocyst stage). Containing several strong anti-oxidants, hormone levels required and a broad spectrum of energy sources.
